Substantially improving the cure rate of high-risk BRCA1-like breast cancer patients with personalized therapy (SUBITO): an international randomized phase III trial

Doelgroep

Klinische of pathologische stadium III mammacarcinoom met eigenschappen van homologe recombinatie deficiëntie (HRD) (dwz BRCA-mutatie, BRCA1 promotor hypermethylatie, BRCA1-like DNAcopy numberprofiel), <65 jaar, geschikt voor autologe stamceltransplantatie.

Onderzoeksopzet

• Fase III

• Internationale, multicenter, gerandomiseerde, open-label, (neo) adjuvante studie

• Interventie: 

  • Arm A: standaard chemotherapie; 4x ddAC, gevolgd door paclitaxel (carboplatin); nadien gevolgd door monotherapie olaparib.
  • Arm B: interventie arm; 4x ddAC; gevolgd door chemotherapie ter voorbereiding op mini stamceltransplantatie.
